Abstract
Infections caused by Staphylococcus aureus remain a significant health concern, particularly due to the increasing of resistance to conventional antibiotics. This situation highlights the need for the development of safer and more effective natural product based antibacterial alternatives, particularly to support the management of skin and soft tissue infections caused by Staphylococcus aureus. Chinese senna (Cassia alata L.) leaves and betel (Piper betle L.) leaves are known to contain active compounds including flavonoids, alkaloids, saponins and tannins which possess potential antibacterial properties. This study aimed to identify the secondary metabolite constituents and evaluate the antibacterial activity of a combination of 96% ethanolic extracts of Chinese senna leaves and betel leaves at a 1:1 ration and concentrations of 25%, 50% dan 75% against Staphylococcus aureus. The study employed a true experimental laboratory design with a post-test only control design. Antibacterial activity was assessed using the disc diffusion method, with 10% Ichthyol ointment serving as the positive control and the diameter of the inhibition zone used ad the observation parameter. The obtained data were analysed using a One-Way ANOVA test to determine differences in antibacterial activity among the treatment groups. The result demonstrated that all extract combinations exhibited antibacterial activity against Staphylococcus aureus within the intermediate category, with the 75% concentration producing the largest inhibition zone. Statictical analysis revealed significant differences among the treatment concentrations (p < 0,05), indicating that extract concentration influenced the resulting antibacterial activity. Based on these findings, the combination of 96% ethanolic extracts of Chinese senna leaves and betel leaves show potential for development as a natural antibacterial alternative against Staphylococcus aureus and may serve as a foundation for further research into the development of natural product based topical formulations.
