Abstract

The untargeted lipidomics approach on beef and pork ham is a method that can be used to provide a lipid profile and has high sensitivity, and has been successfully applied for halal authentication. The objective of this study was to authenticate the halal status of beef and pork ham using Chemoinformatics Coupled Lipidomics Methods. The results showed that the number of sublipids in beef and pork ham was 35 and 26 in negative ions, respectively, while positive ions were 212 and 139. Glycerophospholipids were the lipid class found in both beef and pork ham, in both positive and negative ions. Through PCA analysis, it was found that the lipid profiles of beef and pork ham were different. Cluster analysis using K-Mean could classify information and identify lipid profile patterns in beef and pork ham that were well clustered. PLS-DA analysis of positive ions had an R2 value of 0.99778, while negative ions had an R2 value of 0.99623. This indicates that halal authentication of beef and pork ham is more selective in positive ions. The VIP score results in positive ions showed fifteen potential lipid profiles that can be used for halal authentication, where LPE(18:1)-H is the lipid with the highest composition in beef ham, and LPS(22:6)-H in pork ham. Based on the distinctive lipid profile differences, the results of this study can be used for selective authentication of beef and pork ham.
