Abstract
Red betel leaf (Piper crocatum) is a botanical species that possesses bioactive elements which may serve as natural antioxidants. This research was designed to evaluate IC₅₀ values for DPPH and ABTS methods, to assess the total phenolic content, and to investigate the relationship between antioxidant capacity and total phenolic content.The investigation involved the process of ethanol extraction, followed by the assessment of antioxidant activity using DPPH, ABTS, and FRAP techniques, alongside the measurement of total phenolic content utilizing the Folin Ciocalteu approach. The information collected was examined through linear regression, One-Way ANOVA, Tukey HSD post hoc tests, and pearson correlation assessments. The findings revealed that the IC50 values for both DPPH and ABTS methods were 96.8 ppm (R² = 0.943) and 69.91 ppm (R² = 0.993), respectively, while the FRAP method showed antioxidant activity ranging from 2.77 to 16.19 mg VCE/g extract (R² = 0.975). The statistical evaluation demonstrated that variations in concentration had a significant impact on antioxidant activity (p < 0.05). The total phenolic content exhibited a very strong positive correlation with antioxidant activity, with the most substantial correlation detected in the ABTS method (r = 0.978). In summary, the ethanolic extract of red betel leaf reveals significant antioxidant potential, with the ABTS method recognized as the most sensitive and displaying the strongest correlation with total phenolic content.
